M-CSF, also called CSF1, consists of a 14-amino acid peptide, longer than the usual 8-to-11-mer recognized by most CTLs. The M-CSF gene is mapped to 1p21-p13 and contains 10 exons and 9 introns spanning 20 kb. Although it is a single-copy gene, its expression results in the synthesis of several mRNAs, ranging in size from about 1.5 to 4.5 kb. There are 2 forms of M-CSF, one with 224 amino acids and the other with 522 amino acids, that result from alternative splicing. Furthermore, M-CSF has a role in development of the placenta. Uterine M-CSF concentration is regulated by a synergistic action of estradiol and progesterone. M-CSF is produced by uterine glandular epithelial cells. It had been found that FMS, the M-CSF receptor, is expressed in placenta and choriocarcinoma cell lines. ScienCell's mouse M-CSF ELISA kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. Mouse M-CSF-specific polyclonal antibodies are pre-coated onto 8 x 12 divisible strips. The mouse-specific detection polyclonal antibodies are biotinylated. The test samples and biotinylated detection antibodies are subsequently added to the wells and then washed with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB is used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changes to yellow after adding acidic stop solution. The intensity of yellow is proportional to the amount of mouse M-CSF in the sample that is captured on the strips.
